RESUMO

Konjac species (Amorphophallus spp.) are the only plant species in the world that are rich in a large amount of konjac glucomannan (KGM). These plants are widely cultivated as cash crops in tropical and subtropical countries in Asia, including China. Pectobacterium carotovorum subsp. carotovorum (Pcc) is one of the most destructive bacterial pathogens of konjac. Here, we analyzed the interactions between Pcc and susceptible and resistant konjac species from multiple perspectives. At the transcriptional and metabolic levels, the susceptible species A. konjac and resistant species A. muelleri exhibit similar molecular responses, activating plant hormone signaling pathways and metabolizing defense compounds such as phenylpropanoids and flavonoids to resist infection. Interestingly, we found that Pcc stress can lead to rapid recombination of endophytic microbial communities within a very short period (96 h). Under conditions of bacterial pathogen infection, the relative abundance of most bacterial communities in konjac tissue decreased sharply compared with that in healthy plants, while the relative abundance of some beneficial fungal communities increased significantly. The relative abundance of Cladosporium increased significantly in both kinds of infected konjac compared to that in healthy plants, and the relative abundance in resistant A. muelleri plants was greater than that in susceptible A. konjac plants. Among the isolated cultivable microorganisms, all three strains of Cladosporium strongly inhibited Pcc growth. Our results further elucidate the potential mechanism underlying konjac resistance to Pcc infection, highlighting the important role of endophytic microbial communities in resisting bacterial pathogen infections, especially the more direct role of fungal communities in inhibiting pathogen growth.

Soft rot of konjac (Amorphophallus spp.) is a devastating disease caused by the bacterium Pectobacterium carotovorum subsp. carotovorum (Pcc) with serious adverse effects on plantation development, corm quality and crop yield due to the current lack of effective control measures. The main objective of the present study was to elucidate the mechanisms underlying plant resistance to soft rot disease. A combination of transcriptomic and metabolomic analyses demonstrated significant enrichment of differentially expressed genes (DEG) and differentially accumulated metabolites (DAM) associated with plant hormones, phenylpropanoid biosynthesis and, in particular, alkaloid metabolism, in Amorphophallus muelleri following Pcc infection compared with A. konjac, these data implicate alkaloid metabolism as the dominant mechanism underlying disease resistance of A. muelleri. Quantitative real-time polymerase chain reaction analysis further revealed involvement of PAL, CYP73A16, CCOAOMT1, RBOHD and CDPK20 genes in the response of konjac to Pcc. Analysis of the bacteriostatic activities of total alkaloid from A. muelleri validated the assumption that alkaloid metabolism positively regulates disease resistance of konjac. Our collective results provide a foundation for further research on the resistance mechanisms of konjac against soft rot disease.

This work determined and analyzed the complete chloroplast genome sequence of Amorphophallus konjac K. Koch ex N.E.Br 1858 from Yunnan, China. The genome size was 167,470 bp, of which contains a large single-copy region (LSC 93,443 bp), a small single-copy region (SSC 21,575 bp), and a pair of inverted repeat regions (IR 26,226 bp). The chloroplast genome has 131 genes, including 86 protein-coding genes, 37 tRNAs, and eight rRNAs. A previous study reported deletion of accD, psbE, and trnG-GCC genes in the A. konjac chloroplast genome. Our study supports the conservative structure of A. konjac and does not support the gene deletion mentioned above. Phylogenetic analysis indicated that A. konjac shares a close relationship with another A. konjac (collected from Guizhou) and A. titanium by forming a clade in the genus Amorphophallus. Our results provide some useful information to the evolution of the family Araceae.

The type and content of carbohydrates in konjac corms are an essential factors in determining the quality of konjac; however, the pattern of carbohydrate changes and the mechanism regulating the development of mother and daughter corms in the "relay growth" process of Amorphophallus muelleri remain unclear. This study aimed to investigate changes in corm carbohydrates during the growth cycle of A. muelleri and to compare the carbohydrate composition and the expression of related genes between mother and daughter corms. Integrated metabolome and RNA-seq analyses identified 37 differential metabolites as well as 8074 genes that were differentially expressed between mother and daughter corms, the majority of which were involved in starch and sucrose metabolism. More than 80% of the differential metabolites, including sucrose and starch, tended to accumulate in the mother corms; however, konjac glucomannan (KGM), as one of the most important carbohydrates and its major component of the corm, accumulated in higher amounts in the daughter corms. In addition, the expression of invertase and alpha-amylase that promote the breakdown of sucrose and starch was 351.78- and 15.63-fold higher, respectively, in the daughter corm, whereas that of the starch synthesis gene AkWAXY was only 0.096 times as high as in the mother corms. Furthermore, the level of cellulose synthase-like protein G, which promotes KGM synthesis, was 3.85 times higher in daughter corms compared to mother corms. Thus, we inferred that the daughter and mother corms had two distinct carbohydrate utilization strategies. This study provides insights into temporal changes in carbohydrates during the growth cycle of A. muelleri.

Amorphophallus sp. is an economically important crop for rural revitalization in southwest China. However, Fusarium solani often infects Amorphophallus sp. corms during storage, damaging the corm quality and affecting leaf elongation and flowering in the subsequent crop. In this study, the mechanism of resistance to F. solani was investigated in the leaf bud and flower bud corms of Amorphophallus muelleri through transcriptome and metabolome analyses. A total of 42.52 Gb clean reads and 1,525 metabolites were detected in a total of 12 samples including 3 samples each of disease-free leaf bud corms (LC), leaf bud corms inoculated with F. solani for three days (LD), disease-free flower bud corms (FC), and flower bud corms inoculated with F. solani for three days (FD). Transcriptome, metabolome, and conjoint analyses showed that 'MAPK signal transduction', 'plant-pathogen interaction', 'plant hormone signal transduction', and other secondary metabolite biosynthesis pathways, including 'phenylpropane biosynthesis', 'arachidonic acid metabolism', 'stilbene, diarylheptane and gingerolin biosynthesis', and 'isoquinoline alkaloids biosynthesis', among others, were involved in the defense response of A. muelleri to F. solani. Ultimately, the expression of six genes of interest (AmCDPK20, AmRBOH, AmWRKY33, Am4CL, Am POD and AmCYP73A1) was validated by real-time fluorescence quantitative polymerase chain reaction, and the results indicated that these genes were involved in the response of A. muelleri to F. solani. Ferulic acid inhibited the growth of F. solani, reducing the harm caused by F. solani to A. muelleri corms to a certain extent. Overall, this study lays a strong foundation for further investigation of the interaction between A. muelleri and F. solani, and provides a list of genes for the future breeding of F. solani-resistant A. muelleri cultivars.

Encarsia formosa is a natural enemy of the invasive pest Bemisia tabaci and is known to be a dominant parasitic. The frequency and magnitude of climate extremes, particularly temperature extremes, have increased, which has put insect populations at risk. However, the effects of temperature extremes on E. formosa are not well understood. To examine the impact of short-term extreme temperature exposure on the development and reproduction of E. formosa, eggs, larvae, pupae, and adults were exposed to high/low temperature treatments (HLT25, HLT50, LLT25, and LLT50). Our findings indicate that the pupal stage of E. formosa exhibited the strongest tolerance to both heat and cold, while adults exhibited a weaker tolerance. The shortest egg-to-adult development period of 12.65 days was observed in E. formosa exposed to HLT50 treatment during the egg-larval stage. The parasitism peak of the adult stage was delayed by 1-6 days after exposure to extreme temperatures during the egg-larval stage. Conversely, the parasitism peak was advanced by 1-3 days after exposure to extreme temperatures during the pupal and adult stages. The eclosion rate, total parasitism, eclosion rate of the F1 generation, and adult longevity of the F1 generation were lower in the treatment groups than in the control groups. The F1 generation's development period was prolonged to 15.49 and 15.19 days after exposure to HLT25 and HLT50 treatments, respectively, during the egg-larval stage. The F1 generation's development period was shortened to 13.33 days after exposure to LLT50 treatment during the pupal stage. Male individuals appeared in the F1 generation after exposure to HLT50 treatment during the pupal stage, with females accounting for only 56.38%. Our results demonstrate that short-term exposure to extreme temperatures has detrimental effects on the growth and reproduction of E. formosa. In field biocontrol against E. formosa, the release of E. formosa should be avoided as much as possible when the ambient temperature is higher than 35°C or lower than 0°C. During extreme temperature conditions, timely supplementation and release of E. formosa population, along with ventilation and cooling in greenhouse facilities during summer, are necessary for better pest control efficacy.

Deep neural networks (DNNs) are prone to the notorious catastrophic forgetting problem when learning new tasks incrementally. Class-incremental learning (CIL) is a promising solution to tackle the challenge and learn new classes while not forgetting old ones. Existing CIL approaches adopted stored representative exemplars or complex generative models to achieve good performance. However, storing data from previous tasks causes memory or privacy issues, and the training of generative models is unstable and inefficient. This paper proposes a method based on multi-granularity knowledge distillation and prototype consistency regularization (MDPCR) that performs well even when the previous training data is unavailable. First, we propose to design knowledge distillation losses in the deep feature space to constrain the incremental model trained on the new data. Thereby, multi-granularity is captured from three aspects: by distilling multi-scale self-attentive features, the feature similarity probability, and global features to maximize the retention of previous knowledge, effectively alleviating catastrophic forgetting. Conversely, we preserve the prototype of each old class and employ prototype consistency regularization (PCR) to ensure that the old prototypes and semantically enhanced prototypes produce consistent prediction, which excels in enhancing the robustness of old prototypes and reduces the classification bias. Extensive experiments on three CIL benchmark datasets confirm that MDPCR performs significantly better over exemplar-free methods and outperforms typical exemplar-based approaches.

Coverage path planning (CPP) of multiple Dubins robots has been extensively applied in aerial monitoring, marine exploration, and search and rescue. Existing multi-robot coverage path planning (MCPP) research use exact or heuristic algorithms to address coverage applications. However, several exact algorithms always provide precise area division rather than coverage paths, and heuristic methods face the challenge of balancing accuracy and complexity. This paper focuses on the Dubins MCPP problem of known environments. Firstly, we present an exact Dubins multi-robot coverage path planning (EDM) algorithm based on mixed linear integer programming (MILP). The EDM algorithm searches the entire solution space to obtain the shortest Dubins coverage path. Secondly, a heuristic approximate credit-based Dubins multi-robot coverage path planning (CDM) algorithm is presented, which utilizes the credit model to balance tasks among robots and a tree partition strategy to reduce complexity. Comparison experiments with other exact and approximate algorithms demonstrate that EDM provides the least coverage time in small scenes, and CDM produces a shorter coverage time and less computation time in large scenes. Feasibility experiments demonstrate the applicability of EDM and CDM to a high-fidelity fixed-wing unmanned aerial vehicle (UAV) model.

The emerging event cameras are bio-inspired sensors that can output pixel-level brightness changes at extremely high rates, and event-based visual-inertial odometry (VIO) is widely studied and used in autonomous robots. In this paper, we propose an event-based stereo VIO system, namely ESVIO. Firstly, we present a novel direct event-based VIO method, which fuses events' depth, Time-Surface images, and pre-integrated inertial measurement to estimate the camera motion and inertial measurement unit (IMU) biases in a sliding window non-linear optimization framework, effectively improving the state estimation accuracy and robustness. Secondly, we design an event-inertia semi-joint initialization method, through two steps of event-only initialization and event-inertia initial optimization, to rapidly and accurately solve the initialization parameters of the VIO system, thereby further improving the state estimation accuracy. Based on these two methods, we implement the ESVIO system and evaluate the effectiveness and robustness of ESVIO on various public datasets. The experimental results show that ESVIO achieves good performance in both accuracy and robustness when compared with other state-of-the-art event-based VIO and stereo visual odometry (VO) systems, and, at the same time, with no compromise to real-time performance.

Bemisia tabaci is the main pest of agriculture in many regions of the world. The resistance of whitefly to pesticides has increased as a consequence of the continuous irrational use of wide-spectrum pesticides. Thus, pesticides are no longer always effective as a long-term control method. The agricultural landscape can affect the occurrence of an insect population. The objective of this study was to clarify the occurrence of whitefly and its predators in tomato fields in different agricultural landscapes. Different landscapes are classified into urban, flower, water, and mountain landscapes by the principal component analysis method. In 2018-2019, whitefly had the longest main activity period and the lowest density in the flower landscape. The water landscape helped to maintain the highest densities of whitefly during the main activity period. Nine species of predators were sampled, and Nesidiocoris tenuis, Chrysoperla sinica, Menochilus sexmaculata, and Harmonia axyridis were the dominant species throughout the sampling season in both years. During the main activity period, N. tenuis had the highest density in all sampled landscapes. The density of the dominant predators was the highest in the flower landscape, and each natural predator had the largest temporal niche width in the 2-year sampling period. Bemisia tabaci, N. tenuis, and M. sexmaculata were highly synchronized temporally. The flower landscape showed satisfactory results in suppressing whitefly. Increasing the proportion of flowering plants and increasing the diversity of plant crops in the agricultural landscape can effectively reduce the densities of whitefly during an outbreak.

Agricultural landscape pattern may enhance biocontrol services by supporting parasitoid populations, including parasitoids of Bemisia tabaci Gennadius (Hemiptera: Aleyrodidae). In this study, we selected four landscape types in Yunnan province, in south China, which were characterized by flower fields, mountain, river, and urban areas as their main elements. We then carried out systematic surveys in tomato fields found within each landscape type, to determine the diversity, occurrence, and parasitism rate of parasitoids. We found that parasitoids from the genus Encarsia and Eretmocerus were the main natural enemies present, and the most abundant species were recorded in the flower and the mountain landscapes. Also, Encarsia formosa (Hymenoptera: Chalcidoidea) formed the highest relative abundance regardless of the landscape type. We also found that the population density of B. tabaci in flower landscapes was the lowest than that in other landscape types. During the main activity period of B. tabaci, the parasitism rate in the flower landscape was higher than that in other landscape types. Our findings showed that the occurrence of B. tabaci was relatively mild in the flower landscapes. The landscape type was also beneficial to parasitoids as it enhanced their parasitism rate. Therefore, agriculture environmental schemes should consider increasing the size of flower fields in the surrounding landscape to enhance the sustainable control of B. tabaci by the natural agricultural ecosystem.

Agricultural landscape patterns can affect the population dynamics of pest insects. We selected four landscapes (flower field, mountain, river and urban) based on principal components analysis in Yunnan Province, south China. Through systematic investigation carried out in tomato fields, we intended to clarify the population dynamics and densities of Bemisia tabaci under different landscape types. During the main activity period of B. tabaci, the population densities of B. tabaci nymphs in tomato fields in the river and the urban landscape types were the highest compared to the other landscape types; the population densities of female adults in tomato fields in the river landscape type were also the highest. While the population densities of B. tabaci nymphs and female adults in the flower landscape type, no more than five individuals (ind.) 100 cm-2 leaf in both years, were the lowest. The density of B. tabaci nymphs in the middle position of tomato plants was higher than those in the other positions, while the density of adults in the upper position of tomato plants was higher, regardless of landscape types. Our findings showed that the population growth of B. tabaci can be easily controlled by the flower landscape type.

BACKGROUND: The use of chemical insecticides to control Bemisia tabaci Gennadius (Hemiptera: Aleyrodidae) is widespread, although it might exert a sublethal effect on its dominant parasitoid, Encarsia formosa Gahan (Hymenoptera: Aphelinidae). To investigate the sublethal effect of spirotetramat on E. formosa, we observed the ability of E. formosa to locate and handle the host, oviposit and preen after exposure to sublethal concentrations of spirotetramat. RESULTS: After exposure to spirotetramat at LC50 , the response time of E. formosa to the volatile reached 223.40 s and was significantly prolonged. Only 56.44% of the wasps were attracted by the volatile and the insect crawled the slowest among all of the treatments. The averages of oviposition posture adopted and host handled by each E. formosa in 1 h decreased significantly to 1.79 and 1.27, respectively. At the sublethal concentration of LC10 , 94.59% of the wasps were attracted by the volatile and the insect crawled the fastest. The average of host handled by each E. formosa was 3.92, and the frequency of drumming while walking and drumming the host was 12.34 times per second and 12.30 times per second, respectively, demonstrating a significant acceleration in these abilities. CONCLUSION: These findings demonstrate that spirotetramat induced hormesis in E. formosa on exposure to its LC10 concentration and accelerated its host locating, host handling and frequency of antennae drumming. These findings could assist in balancing the chemical and biological control of B. tabaci and enhancing the efficacy of E. formosa as a biocontrol agent. © 2021 Society of Chemical Industry.

One-shot person Re-identification, which owns one labeled sample among numerous unlabeled data for each identity, is proposed to tackle the problem of the shortage of labeled data. Considering the scenarios without sufficient labeled data, it is very challenging to keep abreast of the performance of the supervised task in which sufficient labeled samples are available. In this paper, we propose a relation-based attention network with hybrid memory, which can make full use of the global information to pay attention to the identity features for model training with the relation-based attention network. Importantly, our specially designed network architecture effectively reduces the interference of environmental noise. Moreover, we propose a hybrid memory to train the one-shot data and unlabeled data in a unified framework, which notably contributes to the performance of person Re-identification. In particular, our designed one-shot feature update mode effectively alleviates the problem of overfitting, which is caused by the lack of supervised information during the training process. Compared with state-of-the-art unsupervised and one-shot algorithms for person Re-identification, our method achieves considerable improvements of 6.7%, 4.6%, and 11.5% on Market-1501, DukeMTMC-reID, and MSMT17 datasets, respectively, and becomes the new state-of-the-art method for one-shot person Re-identification.

Robots are developing in much the same way that personal computers did 40 years ago, and robot operating system is the critical basis. Current robot software is mainly designed for individual robots. We present in this paper the design of micROS, a morphable, intelligent and collective robot operating system for future collective and collaborative robots. We first present the architecture of micROS, including the distributed architecture for collective robot system as a whole and the layered architecture for every single node. We then present the design of autonomous behavior management based on the observe-orient-decide-act cognitive behavior model and the design of collective intelligence including collective perception, collective cognition, collective game and collective dynamics. We also give the design of morphable resource management, which first categorizes robot resources into physical, information, cognitive and social domains, and then achieve morphability based on self-adaptive software technology. We finally deploy micROS on NuBot football robots and achieve significant improvement in real-time performance.

OBJECTIVE: To evaluate the clinical effect of non-penetrating trabecular surgery (NPTS) with mitomycin C (MMC) in primary open-angle glaucoma (POAG). METHODS: Twenty-five eyes of 18 patients with POAG underwent NPTS with MMC during the operation. The mean follow-up was 16.6 months (6 - 28 months). RESULTS: The mean preoperative intraocular pressure (IOP) was (27.93 +/- 7.62) mm Hg (1 mm Hg = 0.133 kPa) which decreased to a mean postoperative IOP of (14.62 +/- 3.53) mm Hg (P < 0.001). After operation, there was an obvious diffuse bleb in each eye, in the 3 months after operation, there were functional blebs in 21 eyes, and the successful filtration was ceased in 4 eyes. The anterior chamber reaction was mild. Hyphema occurred in 1 eye after operation. Intra-operatively, in 4 eyes microperforations occurred in the trabecular meshwork without iris prolapse, and after operation, under the gonioscope, the perforation could be observed in these eyes. In another 2 eyes, there was no intra-operative perforation, but a small fissure was seen by gonioscopy. CONCLUSIONS: NPTS with application of MMC can lower the IOP effectively in this group of patients. The recovery of vision after operation is quick. A few complications occurred after operation. It is a promising technique to treat POAG.
